motif Nucleotide-binding motif type I (7000008)

The nucleotide-binding motif, the so-called Rossmann motif, was first identified in dinucleotide-binding proteins. The nucleotide-binding motif comprises a beta-alpha-beta structural motif in which the first beta-strand and the alpha-helix are connected by a Gly-rich phosphate-binding loop. The loop contains a semi-conserved GX[GS]XX[GSTA] sequence pattern. The first strictly conserved Gly allows a tight turn of the main-chain from the beta-strand to the alpha-helix. The second semi-conserved Gly or Ser permits a close contact of the main-chain to the pyrophosphate of the nucleotide. The third semi-conserved residue allows a close packing of the alpha-helix.
